METHOD OF ANALYSIS OF STP-24
Following tests may be conducted before and after treatment.
After treatment means after 72 Hours of applying STP-24.
TEST 01 - Odour
Deodorization of faecal matter by degradation can be tested by spraying about 250 ml/ 10 sq. m before and after treatment on an open area
Deodorized effect can be observed and it will be found that odor is barely sensible.
That is STEP-24 has decomposed odorous substances very strongly from the offensive
Target.
TEST 02 - Biological Performance
Cultivation test of red chillies using the treated waste water may be conducted.
Untreated After Treatement
50X 100x 150 x 50X 100x 150 x
Height of plants (cm)
Yield per plant (Dry Chillies with 20% moisture)
Results indicate that the treatment improves significantly height of plants and yield of chillies compared to untreated plots..

Lead acetate test:
Dip a strip of paper in lead acetate solution and allow it to dry partially till the strip is moist (do not completely dry).
Take 0.2g sample in a dry test tube and heat till vapours arise. Hold the moist lead acetate paper at the mouth of the test

Black precipitate or colouration on the paper

Test for the above before and after treatment.
TEST 04- Indole
Preparation of Kovac’s reagent:
1. Take a 250ml beaker with 75ml of amyl alcohol.
2. Add 25ml conc. HCl to the amyl alcohol.
3. Dissolve 5g of N, N – Dimethyl – 4 – Amino Benzaldehyde in the above solution.
4. Transfer into a dark brown glass bottle and store at 4oC.

Kovac’s reagent test:
Take 0.2g sample in a test tube + 4ml double distilled universal solvent. Vortex the sample thoroughly. Centrifuge the sample at 16,000 rpm for 30 minutes at 4 degrees C..
Take the supernatant liquid in a fresh test tube and add a few drops of Kovac’s reagent along the sides of the test tube. Let the test tube stand for 15 minutes.

A pinkish-red ring is seen at the junction of the two liquids
|
Indole present
|
Test for Indole before and after the treatment.
TEST 05 - Ammonia:
1. Take 0.2g of sample + 2ml of 0.2N NaOH + Heat the test tube till vapours form.
2. Hold a wet red litmus paper at the mouth of the test tube
> litmus colour changes to blue
> ammonia present
3. Take a glass rod dipped in conc HCl and hold it at the mouth of the heated test tube containing the sample
> dense white fumes of NH4Cl formed
> ammonia present
